Mesenchymal cells isolation from Wharton’s jelly, in perspective to clinical applications
نویسندگان
چکیده
Recent years have seen progress in isolation and expansion of mesenchymal cells (MSCs) from human umbilical cord tissue, namely the Wharton’s jelly. Situated between the subamnion and the perivascular zone that surrounds the blood vessels (Troyer & Weiss, 2008), Wharton’s jelly is a matrix of mucous connective tissue, composed of fibroblast-like stromal cells, collagen fibers and proteoglycans, mainly hyaluronic acid (Can & Karahuseyinoglou, 2007). Several studies have demonstrated the proliferative capacity and multipotentiality of these stromal cells; they were shown to differentiate into a neural phenotype in vitro (Mitchell et al., 2003; Ma et al., 2005; Yang et al., 2005; Lu et al., 2006; Ma et al., 2006; Karahuseyinoglou et al., 2007) and into chondrocytes (Wang et al., 2004; Karahuseyinoglou et al., 2007), osteocytes (Wang et al., 2004; Sarugaser et al., 2005; Conconi et al., 2006; Karahuseyinoglou et al., 2007), cardiomyocytes (Wang et al., 2004), skeletal myocytes (Conconi et al., 2006), adipocytes (Conconi et al., 2006; Lu et al., 2006; Mesenchymal cells isolation from Wharton’s jelly, in perspective to clinical applications

Wharton’s Jelly Mesenchymal Stem Cell: Various Protocols for Isolation and Differentiation of Hepatocyte-Like Cells; Narrative Review
There are several differentiation methods for mesenchymal stem cells (MSCs) into hepatocyte-like cell. Investigators reported various hepatic differentiation protocols such as modifying culturing conditions or using various growth factors/cytokines. In this literature review, we compared different MSCs extraction and isolation protocols from Wharton’s jelly (WJ) and explored various MSCs differ...
متن کاملComparative Analysis of Mesenchymal Stem Cells Isolated from Human Bone Marrow and Wharton’s Jelly
Introduction: Bone marrow (BM) is a known source of mesenchymal stem cells (MSCs) that are used for cell therapy. This study attempts to identify if the Wharton’s Jelly (WJ) is a suitable substitute for BM as a source for MSCs. Materials and Methods: A population of human WJ and BM stem cells were isolated and incubated with fluorescein conjugated antibodies for five specific MSC markers....
متن کاملWharton’s Jelly-derived Mesenchymal Stem Cells can Differentiate into Hepatocyte-like Cells by HepG2 Cell Line Extract
Background: Wharton’s jelly is an unlimited source of stem cells that can be used in cell therapy and tissue engineering without any ethical concern. It has been revealed the cell-free extract could be effective to induce cell differentiation. The objective of this study was to induce Wharton’s jelly-derived mesenchymal stem cells (MSCs) into hepatocyte-like cells by premeabilization of the cel...
متن کاملAssess The Pluripotency of Caprine Umbilical Cord Wharton’s Jelly Mesenchymal Cells By RT-PCR Analysis of Early Transcription Factor Nanog
Objective- In the present study we investigated the isolation protocol, population doubling time (PDT) and the expression of a pluripotential gene by RT-PCR analysis of early transcription factor Nanog in caprine umbilical cord (CUC) Wharton's jelly mesenchymal cells (WJMCs). Design- Experimental in vitro study. Animals- Four mix breed goat. Procedures- CUCs were collected from abattoi...
متن کاملIsolation of Mesenchymal Stem Cells from Wharton’s Jelly in Comparison with Bone Marrow and Their Endothelial Differentiation
Objective: Umbilical cord Wharton’s jelly as source of mesenchymal stem cells (MSCs) can be easily obtained. In this study, MSCs were isolated from bone marrow and Wharton’s jelly and induced to differentiate into endothelial-like cells. Materials and Methods: MSCs were isolated and induced to differentiate into endothelial-like cells using vascular endothelial growth factor (VEGF). The endothe...
